摘要
目的研究葛根素对血管性痴呆(VD)大鼠行为学及额叶细胞凋亡的影响。方法采用大鼠双侧颈总动脉(CCA)永久性阻断法(2-VO)建立VD大鼠模型,大鼠随机分为假手术组、2-2VO组和葛根素组,各组大鼠又分为4、8周2个时间点。用暗回避反应法测定学习记忆能力,并应用免疫组织化学法、原位末端标记法检测大鼠额叶bcl-2蛋白表达水平及凋亡细胞数。结果2-2VO组和葛根素组大鼠潜伏期明显缩短,但与2-2VO组比较葛根素大鼠潜伏期均显著延长。2-2VO组和葛根素组大鼠额叶bcl-2的表达水平及凋亡细胞数均较假手术组明显增高,葛根素组bcl-2的表达水平较2-2VO组明显增高,而凋亡细胞数较2-2VO组明显减少。结论葛根素可通过上调bcl-2基因表达而抑制细胞凋亡,改善VD大鼠的学习记忆能力。
Objective To investigate the effects of puerarin on behavior and apoptosis in frontal lobe in vascular demential rats. Methods Rats of experimental group were treated with a permanent bilateral common carotid arteries (CCA) occlussion(2-VO) to establish vascular demential model:SD Rats were randomly divided into three groups:sham-operated group,2-VO group and puerarin group. The learning-memory ability was evaluated by using passive avoidance test. At corresponding two time points of 4th week and 8th week respectively the dynamic changes on the cell counts of bcl-2 and apoptosis in frontal lobe were determined by immunohistochemical method and TUNEL (terminal deoxynucleotidy and transferase-mediated dUTP-bioton nick end labling), respectively. Results ( 1 )The performance on passive avoidance test of rats in 2-VO group and pueratin group was worse than that of rats in sham-operated group. As compared with the rats in 2-VO group, the performance on passive avoidance test of rats in puerarin group was improved significantly. (:2)In both of 2-VO group and puerarin group, the number of bcl-2 and TUNEL positive cells in frontal lobe were much more than that in sham operated group;the expression of bel-2 in puerarin group was obviously better than that in 2 VO group, while TUNEL positive cells in puerarin group were distictly less than those in 2-VO group. Conclusions Puerarin can inhibit apoptosis through upregulating the expression of Bcl-2 protein in frontal lobe. Puerarin may improve cognitive ability in vascular dementia rats.
