摘要
目的:建立系统化的固相萃取方法。方法:选用SPECMulti-Modal混合相固相萃取柱,建立系统化的固相萃取方法。整个萃取过程包括下列步骤:柱预处理、样品过柱、柱清洗、柱干燥、药物的两步洗脱并收集洗脱液、加入内标以及样品浓缩。对血浆中的21种常见滥用药物进行了固相萃取。结果:对样品和洗脱剂通过固相萃取柱的流速进行了选择优化。样品、酸性洗脱液、碱性洗脱液的最佳流速分别为0.33、1.5和0.5mlmin-1。21种药物在血浆中的绝对回收率都高于86%。
Objective: To develop a systematic SPE extraction method. Methods: The SPEC Multi Modal columns were used to develop a systematic SPE extraction method. The extraction procedure includes 6 steps: column preconditioning, sample addition, column rinse and drying , elution of drugs by two eluents, addition of internal standard and concentration. Twenty one normal abused drugs were extracted from plasma. Results: The effects of the sample and eluent flow rates were investigated, and it was found that the optimal flow rates of sample application,acidic eluent,and basic eluent were 0.33,1.5 and 0.5 ml/min respectively. The absolute recovery rates of 21 drugs from plasma exceeded 86%. Conclusion: The systematic solid phase extraction method can be used for drug screening.
