细粒棘球蚴重组EgP-29诱导免疫小鼠脾细胞增殖及细胞因子水平的变化 被引量:3

摘要 目的探讨细粒棘球蚴重组抗原(rEgP-29)接种小鼠后的免疫保护机制。方法ICR小鼠随机分为免疫组和对照组,采用rEgP-29免疫、Eg原头蚴攻击感染小鼠后20周,以MTT法检测经刀豆蛋白(ConA)和rEgP-29刺激后T淋巴细胞增殖水平,ELISA法检测体外脾细胞诱生的IL-2、IL-4和IFN-γ水平。结果MTT法检测证实rEgP-29免疫组小鼠脾淋巴细胞增殖水平明显高于对照组(P<0.05);ELISA法检测表明,免疫组小鼠经ConA或rEgP-29诱生的IL-2、IFN-γ水平与对照组相比显著增加(P<0.05),而IL-4水平无明显变化。结论rEgP-29可能诱导Th1型免疫应答为主,对抗Eg原头蚴攻击感染。 Objective To investigate the immunologic mechanism in mice immunized with recombinant EgP- 29 vaccine of Echinococcus granulosus against Eg protoscolices. Methods ICR mice were subcutaneously and intranasally vaccinated by the vaccine, followed by the chanllenge with Eg protoscolices on the 8th week of vaccination and the sarifie on the 20th week of infection to separate splenocytes, to dectect the proliferation activity of spleen T lymphocytes by MTT assay, and to measure the level of IL - 2, IL - 4 and IFN -γ by ELISA. Results As demonstrated by the MTT assay, the rEgP- 29 antigen could stimulate specifically the proliferation of splenic T lymphocytes in mice. ELISA assay indicated that the level of IL - 2 and IFN - γ in immunity group were significantly increased after stimulus with ConA or rEgP - 29 ( P 〈 0.05 ). There was no significantly change in the level of IL - 4. Conclusion The rEgP - 29 vaccination induces Thl cellular response in mice against Echinococcus granulosus protoscolices infection.
