紫杉醇联合顺铂治疗晚期食管癌中自噬的研究 被引量:2

摘要 目的:探讨紫杉醇联合顺铂治疗前后,晚期食管癌细胞自噬的表达,为进一步研究食管癌的化疗耐药性提供依据。方法:随机筛选2005年1月~2008年12月郑州大学第二附属医院Ⅲ、Ⅳ期食管鳞癌21例,给予紫杉醇(PTX)加顺铂(DDP)联合化疗1个疗程前后,穿刺活检,使用淋巴细胞分离液分离肿瘤细胞,用电子显微镜观察自噬的表达,用MDC染色检测细胞自噬活性。结果:电镜显示,在化疗处理后的肿瘤细胞胞浆中出现大量的自噬泡(7.86±1.37),对照组未发现自噬泡的生成(0.03±0.28)。MDC染色显示,化疗处理组细胞胞浆中存在大量的MDC标记上的点状结构(27.2±5.6),而对照组中这种结构较少(5.2±1.1)。因此与紫杉醇联合顺铂治疗前相比,化疗后自噬泡显著增多(P<0.05),细胞自噬活性显著增加(P<0.05)。结论:紫杉醇联合顺铂能够明显增加晚期食管癌细胞的自噬活性,这提示抑制自噬,有可能改善食管癌的疗效。 Objective: We observed autophagy in advanced esophageal cancer cells treated with paclitaxel and cisplatin in order to find evidence for chemotherapy resistance. Methods: A total of 21 patients with advanced esophageal cancer were selected. After treatment with paclitaxel (PTX) and cisplatin (DDP) for one course, cancer cells were extracted. The level of autophagy was detected with electron microscopy and MDC staining. Results: According to observations from the electron microscopy images, the number of autophagic vacuoles in advanced esophageal cancer cells was significantly increased after treatment with PTX and DDP. MDC staining showed that the autophagy in these cancer cells was significantly increased after treatment with PTX and DDP. Conclusion: PTX combined with DDP can significantly increase autophagy in advanced esophageal cancer cells, indicating that inhibition of autophagy might improve the therapeutic effects of such treatment for esophageal cancer.
