负载肿瘤抗原的DC疫苗对小鼠体内膀胱肿瘤的抑制作用 被引量:1

摘要 目的研究负载肿瘤抗原的DC疫苗对T739小鼠体内膀胱肿瘤的免疫学效应。方法建立BTT739荷瘤小鼠动物模型,随机分为实验组和实验对照组和空白对照组,于肿瘤细胞接种后第7、14天给予相应DC疫苗治疗,每组分2亚组,分别用于测量瘤质量、体积及用于观察荷瘤小鼠存活情况。结果负载肿瘤抗原的DC疫苗实验组荷瘤鼠肿瘤平均体积和平均瘤质量均显著低于对照组(P<0.01),生存期长于对照组,并且有2只小鼠无瘤长期存活。经DC疫苗实验组治愈的2只小鼠30d后无肿瘤生长,再次皮下接种BTT739细胞观察30d仍无肿瘤发生。结论负载肿瘤抗原的DC疫苗对膀胱肿瘤荷瘤小鼠具有抑瘤效应和生存期延长作用。 Objective To investigate the immunotherapeutic effect of dendritic cells vaccine plus tumor antigen on bladder tumor in T739 mice. Methods After establishment of the B33739 bladder tumor model, 36 T739 mice were randomly divided into experiment group, control group, and empty control group. Peritumoral injection of dendritic cell ( DC ) vaccine was performed on day 7 and day 14 after inoculation of tumor cells. Tumor weight and volume were measured. The life span of mice was also observed. Results The inhib- itory effect of DC vaccine on the tumor weight and volume in the experiment group was significantly higher than that in the control group ( P 〈 0.01 ). The survival time of the mice in the experiment group was significantly longer than that in the control group. Two mice treated with DC vaccine were tumor-free 30 days after being rechallenged with BI3739 cells. Conclusion DC vaccine plus with tumor antigen can inhibit tumor growth and prolong the survival time of T739 mice with BTT739 bladder tumor.
