复合培养对低氧时肺动脉平滑肌细胞表达细胞因子的调节

Effect of cocultured pulmonary microvascular endothelial cells on cytokine expressions in pulmonary arterial smooth muscle cells after hypoxic exposure

摘要 目的研究低氧对肺动脉平滑肌细胞(pulmonary arterial smooth muscle cells,PASMCs)白细胞介素-1β、4、10、13(IL-1β、4、10、13)活性的影响,及复合培养下肺微血管内皮细胞(pulmonary microvascular endothelial cells,PMVECs)对其调控作用。方法大鼠PASMCs单独培养或与PMVECs复合培养,分为正常组、低氧作用2h(H2)、6h(H6)、12h(H12)、24h(H24)组,采用ELISA法测定各组培养细胞上清中IL-1β、IL-4、IL-10、IL-13的活性。结果低氧刺激后PASMCs的IL-1β、IL-4、IL-10、IL-13活性2h开始升高,6h达高峰,12h降低;各时相点复合培养组均低于对应的单独培养组(P<0.01)。结论低氧可以激活PASMCs的IL-1β、IL-4、IL-10、IL-13活性,在复合培养条件下,PMVECs对此效应具有下调性影响。 Objective To study the effect of hypoxia on the expressions of interleukin-1β, 4, 10, and 13 (IL-1β, IL-4, IL-10 and IL-13) in pulmonary arterial smooth muscle cells (PASMCs) and the regulative role of cocultured pulmonary microvascular endothelial cells (PMVECs). Methods Rat PASMCs were eocuhured with or without rat PMVECs, and randomly divided into 5 groups, normal group (N), 2-hour hypoxia (H2), 6-hour hypoxia ( H6), 12-hour hypoxia ( H12), and 24-hour hypoxia (H24). The expressions of IL-1β, IL-4, IL-10 and IL-13 in the supernatant were detected with ELISA. Results The expression levels of IL-1β, IL-4, IL-IO and IL-13 was increased in H2 group, peaked in H6 group, and then decreased in HI2 group. However, the expression levels of above-mentioned cytokines in eocuhured groups were significantly lower than those in corresponding hypoxia group. Conclusion Hypoxia can enhance activities of 1L-1β, IL-4, IL-10, and IL-13 in PASMCs, but this enhancement can be down-regulated by cocultured PMVECs.
