高效液相色谱法测定导赤散及其配伍组中马兜铃酸A的含量

Detection on contents of aristolochic acid A in Daochi powder and its combination group with high performance liquid chromatography

目的通过比较导赤散及其配伍组中马兜铃酸A的含量,探索方剂配伍减毒的作用。方法采用色谱法,色谱柱为D iamon TmC18(250mm×4.6mm,5μm),甲醇-水-冰乙酸(72∶27∶1)为流动相;柱温:室温;检测波长315nm;流速1.0mL/m in。结果关木通组、导赤散组、配伍一组、配伍二组的马兜铃酸A含量分别为:0.549mg/g、0.357mg/g、0.226mg/g、0.316mg/g。结论生药关木通经过配伍后,马兜铃酸A的含量能显著降低。

Objective To explore the toxitcity - reducing effect in prescription compatibility via comparison on content of aristolochic acid A between Daochi powder and its components group. Methods Chromatography was applied in this study, with Diamon TmC18 （250mm ×4.6mm, 5μm ） used as chromatographic column, methanol -water-glacial acetic acid （72：27：1） as mobile phase and room temperature selected as the column temperature, a parameter 315 nm as the detection wavelength and a 1.0mL/min as the velocity. Results Contents of aristolechic acid A were detected as： 0.549mg/g in Guanmntong （caulis hocquartiac） group, 0.357mg/g in Daochi powder group, 0.226mg/g in combination group one and 0.316mg/g in combination group 2. Conclusion Content of aristoleehic acid A declined notably after the crude caulis hocquartiae was combinafively used.
