Speciation of magnesium in rat plasma using capillary electrophoresis inductively coupled plasma atomic emission spectrometry

摘要 研究毛细管电泳-电感耦合等离子体原子发射光谱联用技术应用于大鼠血浆中镁元素形态分析的新方法,并对游离态的镁进行表征.实验优化条件为:融熔毛细管120 cm×100μm(I.D.);缓冲溶液50 mmol/LNaAc-HAc(pH=5.5);电泳电压20 kV.在该条件分析大鼠血浆样品中Mg的形态主要有7种,通过标准Mg的迁移时间及加标后迁移时间定位表征血浆样品中游离镁的形态峰,各形态迁移时间RSD<3%,峰面积定量血浆样品中游离镁的含量为14.0 mg/L,RSD<5%.采用以上联用技术对大鼠血浆样品中游离镁进行了加标回收测定,其加标回收率为95.8%-103.8%. A new method for speciation analysis of magnesium species and quantification of free magnesium concentrations in rat plasma was developed by CE-ICP-AES. Baseline separation of magnesium speciation was achieved by using fused-silica capillary, a 20 kV separation voltage and a solution of 50 mmol/L NaAc-HAc (pH=5. 5) as electrolyte buffer. CE-ICP-AES analysis of a rat plasma sample showed the presence of seven magnesium species, one of which was identified as free Mg^2+ ion. The concentration of free magnesium in the plasma was 14.0 mg/L. RSD of migration time and peak areas were less than 5%. The recoveries of the free magnesium species in four samples ranged from 95. 8 to 103. 8%.
