摘要 目的通过比较酵母膏与酵母膏+乙胺丁醇灌胃致小鼠血清尿酸水平的变化,探讨建立小鼠高尿酸血症模型新方法。方法 52只雄性昆明小鼠,完全随机分为对照组、酵母膏组、酵母膏+乙胺丁醇组及别嘌呤醇组4组,分别采用蒸馏水、酵母膏、酵母膏+乙胺丁醇、酵母膏+乙胺丁醇+别嘌呤醇片灌胃,3周后取血测定小鼠血清尿酸水平。结果连续灌胃3周后,各组小鼠血清尿酸水平分别为:酵母膏组(247±61)μmoL/L,酵母膏+乙胺丁醇组(30l±53)肛mol/L、对照组小鼠血清尿酸为(235±34)肛moL/L,别嘌呤醇组血清尿酸(114.67±49.74)μmoL/L。结论酵母膏+乙胺丁醇灌胃可建立相对持久稳定的小鼠高尿酸血症模型,同时升高的血清尿酸可以被阳性药物别嘌呤醇片有效降低。 Objective To explore a new method of establishing a mouse model of hyperuricemia by intragastric administration of yeast extract and yeast extract plus ethambutol. Methods A total of 52 male Knnming mice were randomly divided into 4 groups, control group, yeast extract group, yeast extract plus ethambutol group and allopurinol group. Intragastric injection of distilled water, yeast extract, yeast extract plus ethambutol, and yeast extract combined with ethambutol and allopurinol was administrated in each group respectively for 3 weeks. Then the concentrations of serum uric acid were measured. Results The concentrations of serum uric acid in control group, yeast extract group, yeast extract plus ethambutol group and allopurinol group were ( 235±34 ) μmol/L, ( 247±61 ) μmol/L, ( 301 ± 53 )μmol/L and ( 114. 67 ±49. 74) μmol/L respectively. The level of serum uric acid was higher in yeast extract plus ethambutol group than in yeast extract group and control group ( both P 〈 0. 05 ). The concentration of serum uric acid in allopurinol group was lower than in yeast extract group and yeast extract plus ethambutol group ( both P 〈 0. 05 ). Conclusion Intragastric administration of yeast extract plus ethambutol is a good method to establish a stable and lasting mouse model of hyperuricemia. Allopurinol can effectively lower the level of serum uric acid in such a mouse model of hyperuricemia.
