摘要
目的:研究持续运动后大鼠杏仁核细胞内钙浓度的时程变化。方法:使用钙荧光指示剂Fura-2/AM负载大鼠脑杏仁核神经元细胞,用荧光双波长分光光度计比值法测定持续游泳运动后各时段及对照组的胞浆游离钙浓度。结果:当细胞外钙离子浓度为1.0 mmol/h,对照组钙离子浓度为362.12±24.30nmol/L,运动后即刻组钙离子浓度为388.23±17.40nmol/L,1h组钙离子浓度为89.70±14.50nmol/L,2h组钙离子浓度为278.31±15.20nmol/L,4h组钙离子浓度为322.26±19.30nmol/L。各组间数据存在显著差异(P<0.01)。结论:持续运动后大鼠脑杏仁核神经细胞内游离钙离子浓度总体呈先升后降,再升复常的时程变化。
Objective: To study the time interval change of intracellular free calcium density in nucleus amygdalate neuron cells after continued swimming exercise. Methods: Nucleus amygdalate neuron ceils are loaded with calcium fluorescent indicator Fura -2/AM, and intracellular free calcium density in each group is measured by using Dual - wavelength fluorescence spectrophotometer ratio method. Results: On the condition that the extra cellular calcium density is 1.0 mmol/1, intracellular free calcium density in the control group is 362.12 ±24.3nmol/L, 388.23 ± 17.40nmob/L in the instantly group, 89.70 ± 14.50nmol/L in the lh group, 278.31 ± 15.20nmol/L in the 2h group and 322.26 ± 19.30nmol/L in the 4h group. There is marked difference in each group index( P 〈 0.01 ) . Conclusion: The time interval change of intracellular free calcium density in nucleus amy plays the general trend of rising first, then falling and finally gdalate neuron ceils after continued swimming exercise distuming normal.
