穿心莲指纹图谱和不同产地质量研究 被引量:12

摘要 目的:建立穿心莲药材有效的质量评价方法。方法:通过建立HPLC指纹图谱和有效成分的含量测定,对来自10个不同产地的穿心莲药材进行分析。色谱条件为Alltima C18色谱柱(4.6mm×250mm,5μm),流动相为乙腈-水系统,梯度洗脱,流速1.0mL.min-1,指纹图谱检测波长为215nm,穿心莲内酯和脱水穿心莲内酯含量测定的检测波长分别为225nm和254nm。结果:得到分离度、重现性均较好的穿心莲药材HPLC指纹图谱,标示了10个共有色谱峰,对10批来自不同产地穿心莲样品的色谱指纹图谱进行了相似度评价;应用HPLC-DAD测定了不同产地穿心莲样品中穿心莲内酯和脱水穿心莲内酯的含量,结果显示,穿心莲药材中穿心莲内酯和脱水穿心莲内酯总量介于1.09%~4.47%。结论:该方法简便准确,灵敏度高、重复性好,可以作为穿心莲药材的质量控制方法,此方法已被《香港中药材标准》采用。建议HPLC指纹图谱用于穿心莲药材的质量控制,穿心莲内酯和脱水穿心莲内酯的总量不得低于1.0%。 Objective: To establish an effective method to assess the quality of Herba Andrographis. Methods: fingerprinting and content determination methods were applied to evaluate ten batches of Herba Andrographis. The samples were separated by an Alhima C18 column (4. 6mm × 250mm, 5μm) using acetonitrile -water gradient system as mobile phase. The flow rate was 1.0mL · min^-1 and the wavelengths for content determination of androgra , the detection wavelength for fingerprinting was at 215 nm, pholide and dehydroandrographolide were at 225 arid 254 nm respectively. Results: HPLC fingerprint of Herba Andrographis was established with good separation and repeatability which could be used for quality assessment of Herba Andrographis. Ten common peaks were defined in characteristic fingerprints, and Similarity ten batches of Herba Andrographis, the total content of Evaluation system was applied to evaluate fingerprint of the andrographolide and dehydroandrographolide was between 1.09% and 4. 47%. Conclusion: The method is sensitive, repeatable and accurate, which can be used as quality control for Herba Andrographis and has been used in Hong Kong Chinese Materia Medica Standards. We suggest that the HPLC fingerprinting should be involved in the quality control of Herba Andrographis, and the total content of andrographolide and dehydroandrographolide in Herba Andrograohis is not less than 1.0%.
