摘要
提出了超高效液相色谱法同时测定红景天保健品中红景天苷和甙元酪醇含量的方法。样品经甲醇超声提取,以Acquity UPLC^TM BEH色谱柱(2.1mm×50mm,1.7μm)为固定相,以磷酸(0.05+99.5)溶液-乙腈混合液为流动相作梯度淋洗,用二极管列阵检测器,检测波长为221.3nm。红景天苷和甙元酪醇的质量在0.01~0.3g·L^-1范围内与其吸光度呈线性关系。应用此法测定红景天茎根和红景天胶囊,平均回收率在90.67%~95.55%和90.73%~96.52%之间。
A method for simultaneous determination of salidroside and tyrosol in health-care food of rhodiola by Ultra-HPLC was proposed. The sample was extracted with methanol, and separated on Acquity UPLC^TM BEH column (2.1mm×50mm,1.7μm), using mixed solution of phosphoric acid (0. 05+99. 5) and acetonitrile with different ratio as mobile phase in gradient elution. Photodiode array detection was made at the wavelength of 221.3 nm. Linear relationships between the peak areas and mass of salidroside and tyrosol were obtained in the range of 0. 01- 0. 3 g·L^-1. The proposed method was used in the analysis of root of rhodiola and capsule of rhodiola, values of recovery found by standard addition method were in the range of 90. 67%- 95. 55% and 90. 73%--96. 52% respectively.
