鹅肥肝中差异表达基因的筛选、克隆及分析 被引量:5

摘要 实验以12只朗德鹅为实验素材,采用mRNA差异显示技术检测填肥组和正常组鹅肝中的差异表达基因。实验结果显示,共获得22条差异表达条带;对其中1条差异表达极显著的片段测序和分析,获得该基因部分mRNA序列731bp,与鸡和人纤维蛋白原γ(FGG)基因同源性分别为88.78%和76.91%。进一步半定量RT-PCR检测发现,该基因在对照组中表达水平极显著高于填肥组(P<0.01)。该基因在鹅肝中的特异性下调控表达暗示其在鹅脂肪肝形成和耐受中发挥作用,是鹅肝脏脂类代谢研究和肥肝品系选育中重要的候选基因。 Twelve Landes geese were divided randomly into overfeeding group and control group and 6 geese pergroup. The differential expression bands between two groups were detected with the technique of mRNA differential display, and 22 differential expression bands were obtained. A band which displayed significant differential expression between control group and overfeeding group was chosen to clone, check and analysis. The result showed that the fragment has highly homology to human fibrinogen gamma chain gene (FGG) found from The 731 bp sequence (GenBank Accession No.EU326492), which share 88.78% and 76.91% homology to duck's and human's FGG mRNA sequences, respectively. Semi-quantitative RT-PCR revealed that the expression level of geese FGG mRNA is significantly higher in control group than overfeeding group. The special down-regulation of FGG gene implied it may be an important factor in the formation and tolerance of fatty liver in the goose.
