摘要
制备高质量骨关节组织的石蜡切片,是组织病理学评价相关动物骨关节炎的重要技术支撑。通过在兔一侧膝关节的股骨内侧髁关节面制造直径3mm深度4mm的全层关节软骨缺损,获得兔骨关节炎动物模型。术后4周,分离股骨髁,经过10%中性甲醛固定和混合酸脱钙后制备组织石蜡切片,并行苏木精-伊红(HE)和番红-固绿染色。结果显示:造模侧损伤填充区细胞表现为软骨样细胞特征。该法制备的骨关节组织切片,标本平坦完整,软骨和骨组织结构分明。是一种快速有效的骨关节组织石蜡切片的制备方法,为骨关节炎相关动物的组织病理学评价奠定基础。
To provide an essential technique.in evaluation of osteoarthritis animal models, we established a rapid and reliable method of tissue preparation for articular joint histology. A full-thickness three-millimeter-diameter defect extending from the surface of the cartilage to the subchondral marrow space was created on the medial femoral condyle of rabbit knee using a stainless-steel electric drill. The entire femoral condyles of the knees at 4 weeks after the operation were fixed in 10% neutral-buffered formalin and decalci- fied in the mixed acid decalcifying solution. The tissue sections were stained with hematoxylin-eosin (HE) and safranin-O, and observed under a microscope. Histological evaluation of the tissue sections revealed that the newly regenerated cartilage in the fullthickness cartilage defect were cartilage-like cells. Sections prepared with the modified method, were even, integrated, and structure of the bone and cartilage was clear. The technique provided a reliable method for preparation of paraffin section of articular joint, which should facilitate the pathological evaluation of rabbit osteoarthritis models.
