竹黄产胞外多糖液态培养条件优化

摘要 目的:优化竹黄Shiraiasp.SUPER-H168产胞外多糖的液态培养基组成和发酵条件。方法:通过单因素和正交试验优化培养基组成和发酵条件,在65L气升式发酵罐中进行验证试验。结果:经分析确定最佳培养基组成为(g/L)葡萄糖40,玉米浆粉3,KH2PO41,MgSO4.7H2O0.05;最佳发酵条件为起始pH6.0,温度30℃,装液量75mL/250mL,接种量10%(v/v)。在此条件下,摇瓶中生物量及多糖分别达到11.7g/L和1.12g/L,验证试验中分别可达到12.1g/L和1.35g/L。结论:放大试验结果优于摇瓶水平,实验数据对生产有一定指导作用。 Objective: The optimal submerged culture medium and fermentation conditions of exopolysaccharides (EPS) produced by Shiraia sp. SUPER- H168 were obtained. Method: Single factor experiment and orthogonal experiment were conducted to optimize submerged culture medium and fementation conditions. And the scale - up experiments was conducted by 65L air-lift fermentor. Result: The optimal fermentation medium obtained by orthogonal tests contained (g/L) glucose 40, corn steep powder 3, KH2PO4 1, MgSO4· 7H2O0. 05. The optimal fermentation conditions were obtained as follows: initial pH 6.0, fermentation temperature 30℃, 75mL liquid medium in 250mL sized flask, inoculation amount was 5 % ( V/ V). Under the optimal conditions, the biomass and the content of EPS were 11.7g/L and 1.12g/L in flask - shaking experiments, 12. 1g/L and 1.35g/L in the scale - up experiments. Conclusion: The results of scale - up experiments were better than those of flask - shaking experiments under the same operating conditions.
