摘要 目的:分离培养胎儿骨髓间充质干细胞(fetus mesenchymal stem cells,fMSCs),分析其基本的生物学特征,如形态特征、增殖特性、细胞表型和多向分化潜能以及核型分析等,为其临床应用提供理论依据。方法:淋巴细胞分离液分离胎儿骨髓MSCs,在相同条件下分别考察各代细胞形态、生长、表面标记的变化情况,并进行不同生长时期的核型分析、细胞周期检测等。结果:在扩增过程中,MSCs的增殖能力随着代数增加而降低。在扩增至第3代时,MSCs表现出较高的纯度,表达CD29、CD44、CD105、CD166,不表达CD34、CD45。10代之前核型未发生变异,仍具成骨成脂分化能力,细胞周期分析显示其仍具有干细胞特性。结论:在体外培养条件下,10代以前的细胞生长性状稳定,可作为组织工程等临床应用的良好对象。 Objective To investigate the biological characters of fetus mesenchymal stem cells (fMSCs) in vitro, such as morphological and proliferative characteristics, phenotype, multi-directional differentiation potential, and karyotype. Methods fMSCs were isolated by density gradient centrifugation from lydroxypropylmethyl cellulose. Of each passage cells, morphology, growth, surface markers, and cycle were investigated. Karyotype were analyzed in different passage cells. Results The proliferative ability of fMSCs was decreased during culture expansion. CD29, CD44, CD105, CD166 were expressed, and CD34, CD45 were not expressed in fMSCs amplified to the 3rd passage. Karyotype of fMSCs was normal before passage 10. Conclusion The state of fMSCs before the 10th passage is stable, and can be a valuable subject for basic research and clinical application.
