摘要
目的:探讨草苁蓉(BR)对大鼠肝脏化学致癌作用的抑制效应。方法:按SoltFarber氏方法复制大鼠肝脏癌前病变模型,用免疫组织化学方法观察了在肝脏癌前病变组和BR处理组及正常对照组大鼠肝脏中胎盘型谷胱甘肽S转移酶(GSTP),突变型P53蛋白及P21蛋白的表达水平。结果:发现500mg/kg剂量的BR甲醇提取物对二乙基亚硝胺(DEN)诱发F344大鼠肝脏GSTP阳性灶的形成有抑制作用,而且其突变型P53蛋白及ras基因产物p21蛋白的表达水平显著低于癌前病变组,并在BR处理后,在肝脏化学致癌初期由于癌前病变的形成所增高的γGT活力明显下降。结论:提示BR对DEN诱发大鼠肝脏癌前病变有抑制作用。
Objective: To investigate the inhibitory effect of Boschniakia rossica(BR)on hepatocarcinogenesis in rats. Method:Based on immunohistochemistry techniques,the expression of placental form glutathione Stransferase(GSTP),mutant p53 and p21 protein were investigated in hepatic preneoplastic lesions induced by SoltFarber protocol in the liver of rats that had been treated with the above method, administered with BR extract and of control groupResult:The extract of BR(500mg/kg)has inhibitory effect on the formation of diethylnitrosamineinduced GSTPpositive foci in F344 rat and the expression of mutant p53 and p21 protein was lower than that of hepatic preneoplastic lesions, and the increasing glutamyltranspeptidase(GT)activity in rat liver treated with SoltFarber protocol was decreased by the extract of BR,Conclusion:These results indicate that BR has inhibitory effect on DEN induced hepatic preneoplastic lesions in F344 rat
