摘要
目的:利用RP-HPLC法测定5种发酵虫草制剂百令胶囊、宁心宝胶囊、心肝宝胶囊、至灵胶囊及金水宝胶囊中3种核苷类成分尿苷、鸟苷及腺苷及2种碱基成分尿嘧啶、腺嘌呤的含量,并分析不同制剂中这5种成分的差异。方法:采用HPLC法测定了28批发酵虫草制剂中尿苷、鸟苷、腺苷及尿嘧啶、腺嘌呤5种成分的含量,并应用SPSS for Windows统计分析软件中的系统聚类分析法对样品中5种成分的信息进行分析。色谱柱为Lichrospher C18柱(250mm×4.6mm,5μm),检测波长260nm,流动相为甲醇-0.05moL·L-1磷酸二氢钾溶液(8∶92),流速1.0mL·min-1。结果:28批发酵虫草制剂中5种成分含量,尿嘧啶为0.03~0.62mg·g-1,尿苷为1.33~8.59mg·g-1,腺嘌呤为0.02~0.53mg·g-1,鸟苷为0.89~6.04mg·g-1,腺苷为1.11~5.78mg·g-1。应用SPSS for Windows统计分析软件中的系统聚类分析法(组平均法)对发酵虫草制剂的质量进行了计算机快速鉴别分类,结果表明3批百令胶囊样品与其他4种制剂25批样品的类距离最远,提示百令胶囊与其他4种制剂的差异最大。3批金水宝胶囊样品聚为一类,9批心肝宝胶囊样品聚为一类,但金水宝胶囊与心肝宝胶囊二者的类距离较小,说明二者的差异也较小。而10批宁心宝胶囊样品和3批至灵胶囊样品合聚为一类,说明二者没有明显的差异。结论:本法操作简便、灵敏,能够较科学、客观、综合地评价发酵虫草制剂的质量。
Objective:To develop the method for the determination of contents of three nucleosides (uridine, guanosine, and adenosine) and two bases (uracil and adenine) and study the difference of chemical constituents in preparations of fermental Cordyceps ( Bailing capsules, Ningxinbao capsules, Xinganbao capsules, Zhiling capsules and Jinshuibao capsules). Method: Twenty - eight preparations of fermental Cordyceps were collected and five constituents( uridine, guanosine, adenosine, uracil, and adenine) in them were determined by RP- HPLC method. Meanwhile, the datas of whole chemical constituents of 28 samples were obtained from the chromatograms and analyzed by Hierarchical Cluster method. The analytes were carried out on a Lichrospher C18 column (250 mm × 4. 6 mm,5 m) and mobile phase consisted of methanol - 0.05 mol · L^- 1 potassium dihydrogen phosphate ( 8 : 92).The flow rate was 1.0 mL · min^-1 and the detection wavelength was set up at 260 nm. Results: The contents of 5 constituents in 28 preparations of fermental Cordyceps were 0. 03 - 0. 62 mg· g^-1 for uracil, 1.33 - 8.59 mg· g^-1 for uridine,0. 02 -0.53 mg·g^-1 for adenine,0. 89 -6.04 mg· g^-1 for guanosine and 1.11 -5.78 mg·g^-1 for adenosine. To classify the preparation of fermental Cordyceps with Hierarchical Cluster method by computer. Three samples of Bailing capsules were classified from the other samples. Three samples of Jinshuibao capsules were clustered together. Nine samples of Xinganbao capsules were clustered together. They were similar. Ten samples of Ningxinbao capsulos and 3 samples of Zhiling capsules were clustered together. They were not absolutely different. Conclusion: The method is effective, sensitive and accurate as well as specific and can assess the quality of preparations of fermental Cordyceps comprehensively.
