应用VNTR技术对吉林省结核分枝杆菌基因分型的研究 被引量:4

摘要 目的:探讨VNTR(variable number of tandem repeat)技术在吉林省结核分枝杆菌基因分型中的应用。方法:初步选取15个分型效果较好的VNTR基因位点,采用聚合酶链反应(PCR)和琼脂糖凝胶电泳,并应用BioNumerics(Version3.0)软件进行处理,建立检测结核分枝杆菌DNA指纹多态性的方法,分析耐药结核分枝杆菌的DNA多态性。结果:共对320株结核分枝杆菌的15个VNTR位点进行了检测,根据这些菌株的指纹多态性特征,共分成26个型别,其中大部分属于一个型(Ⅰ型),占82.5%(264/320),其他菌株呈散在分布。结论:资料分析表明,吉林省结核分枝杆菌的传播似以Ⅰ型菌株为主,应加强此型菌株流行的监控。 Objective:To discuss the application of VNTR( variable number tandem repeat)in the genotyping of Mycobacterium tuberculosis in Jilin Province. Methods:The initial 15 better- performing VNTR gene sites were selected, using polymerase chain reaction (PCR) and agarose gel electrophoresis, and BioNumerics (Version3.0) software to process, the Mycobacterium tubercu- losis DNA Polymorphism fingerprinting methods is established and the resistance of Mycobacterium tuberculosis DNA polymorphism are analyzed. Results: A total of 320 Mycobacterium tuberculosis VNTR of 15 sites were determined, in accordance with these strains polymorphism of the fingerprints are divided into 26 types, most of which belong to one type ( Ⅰ type), accounting for 82. 5% (264/320), other strains were of scattered distribution. Conclusion:All materials show that the type Ⅰ genotype is the main epidemic strain in Jilin Province and we should strengthen the monitoring of the epidemic strain.
