砷铈催化酶标仪比色法测定尿碘 被引量:3

Determination of urinary iodine by cerium catalyzed arsenic reaction and microplate reader colorimetric assay

摘要 目的:用酶标仪代替分光光度计比色提高尿碘检测速度及每次检测样品量。方法:用酶标反应板条代替比色杯,多道移液器加检测试剂,用酶标仪于405 nm+630 nm双波长比色测定。结果:方法的标准曲线线性范围碘质量浓度在0~300μg/L,标准曲线的平均相关系数为-0.9992;精密度:测定尿碘质量浓度为55.6、134.1、201.7μg/L时,其相对标准差(RSD)分别为4.6%、3.2%、1.4%(n=6);准确度:测定碘质量浓度为64.7、231μg/L的尿碘国家标准物质,测定结果与标准物质给定值的相对偏差(RD)分别为2.1%和-1.7%(n=6)。结论:该方法的标准曲线线性关系、精密度、准确度均能满足要求,大大地提高了工作效率,减少了试剂用量,每次检测的最大样品数是行标法的12倍。 Objective:Microplate reader replaces the spectrophotometer colorimetric to improving the analyzes speed and each time the urine iodine analyzes sample quantity. Results : Methods of standard curve linear range of iodine concentration in the 0 - 300 μg/L, standard curve average correlation coefficient of - 0. 9992, precision: analyzes the quality concentration of urinary iodine 55.6,134. 1, 201.7 μg/L, the relative standard deviation (RSD) respectively were 4.6% , 3.2% , 1.4% ( n = 6) , accuracy : analyzes the iodine concentration was 64.7,231 μg/L national standard of urinary iodine material, measurements and standards were given the value of relative deviation (RD) respectively were 2. 1% and - 1.7% (n = 6). Conclusion: This method standard curve lineafity relations, precision and accuracy can meet the requirements,, improve the working efficiency greatly, reduced the reagent consumption, each time analyzes the biggest sample quantity is standard method 12 times.
