摘要
目的:利用生物信息技术,对丙型肝炎病毒NS5区抗原决定簇进行预测分析,获得具有良好抗原的重组蛋白。方法:以Goldkey程序,通过综合亲水参数、可及性参数、柔韧性参数、抗原性参数、氨基酸序列的电荷分布和β折叠等多种指标,对HCVNS5区抗原决定簇进行预测分析,克隆并表达部分NS5区基因。结果:发现7个可能性最大的抗原决定簇肽段,在此基础上表达的NS5区融合蛋白可与丙型肝炎病人血清发生特异性反应。结论:根据Goldkey程序的预测结果。
Objective: To predict epitopes of NS5 protein in hepatitis C virus and to obtain recombinant protein with strong antigenicity. Methods: Epitopes of NS5 protein was predicted and analyzed with Goldkey program, by synthesizing a number indexes including hydrophilicity (Hoop Woods), accessibility, flexibility, charge of amino acid and β turn. Partial NS5 gene was cloned and expressed in E.coli. Results: Seven dominant epitopes of NS5 protein were obtained by prediction, and expressed recombinant protein showed specific reactivity with sera from hepatitis C patients. Conclusion: According to the predicting results, recombinant protein of HCV NS5 with strong antigenicity was obtained.
