摘要
目的:在原代培养的小鼠肝细胞上观察白细胞介素1β(IL1β)对醋氨酚诱导的细胞损伤的影响及其与一氧化氮、环磷酸鸟苷(cGMP)的关系。方法:以两步灌流法分离小鼠肝细胞,进行原代培养;以放射免疫法测定细胞内cGMP和环磷酸腺苷(cAMP)含量。结果:IL1β可减轻醋氨酚诱导的转氨酶漏出,增加肝细胞内cGMP含量,一氧化氮合酶阻断剂LNMMA可完全阻断IL1β的抗损伤作用。结论:IL1β对醋氨酚诱导的肝细胞损伤具有直接的保护作用,这一作用可能是通过一氧化氮、cGMP介导的。
Objective: To investigate the effect of interleukin1β on acetaminopheninduced cell injury and its relation to nitric oxide and cGMP in primary culture of mouse hepatocytes. Methods: Mouse hepatocytes were isolated by two-step perfusion and cultured primarily. cGMP and cAMP contents were measured by radioimmunoassay. Results: Interleukin1β could reduce acetaminopheninduced leakage of aminotransferases and increase the content of cGMP in hepatocytes. LNMMA, an inhibitor of nitric oxide synthase, could completely abolish the antiinjury effect of interleukin1β. Conclusion: Interleukin1β had direct protective effect on acetaminopheninduced injury in hepatocytes and this effect might be mediated by nitric oxide and cGMP.
