实验性肝纤维化过程中Ⅳ型胶原酶活性的研究

TYPE IV COLLAGENASES ACTIVITY IN THE PROCESSES OF EXPERIMENTAL LIVER FIBROSIS

目的了解实验性肝纤维化过程中Ⅳ型胶原降解酶活性的动态变化。方法以人血白蛋白免疫损伤性大鼠肝纤维化为模型；检测实验性肝纤维化各阶段肝组织及血清Ⅳ型胶原降解酶活性的变化。结果肝组织活化及潜在Ⅳ型胶原降解酶活性在致敏阶段无明显变化；在尾静脉攻击中期显著升高；在尾静脉攻击后较前明显下降；在造模后１．５月及３个月时，潜在Ⅳ型胶原酶活性仍显著高正常对照组。血清Ⅳ型胶原降解酶活性与肝组织潜在Ⅳ型胶原降解酶活性呈显著正相关。结论血清Ⅳ型胶原降酶活性的变化反映了肝组织Ⅳ型胶原降解酶活性的改变。

Objective To investigate the alteration of type IV collage-degrading enzyme activity in the processes of experimental liver fibrosis, Methods Immune complex induced animal model of liver fibrosis was developed in Wistar rats. Active and latent hepatic type IV collagen- degrading enzyme activity was measured. Results Active and latent hepatic type IV collagen- degrading enzyzme activity was not altered significantly during immunizing period and were increased remarkably in the middle of intravenous booster period. Then, they declined gradually. But three months after the model production, latent hepatic type IV collagen-degrading enzyme activity was still much higher than that of normal control groups. Serum type IV collagen-degrading enzyme activity was correlated significantly with hepatic type IV collagen-degrading enzyme activity. Conclusion Hepatic type IV collagen-degrading enzyme activity rose significantly in the processes of fibrogenesis, and decreased in the stage of liver cirrhosis and maintained at relatively high levels during reversal of liver cirrhosis. Alterations of serum type IV collagen-degradling enzyme activity could reflect the changes of hepatic type IV collagen-degrading enzyme activity. [
