双波长紫外分光光度法测定生物转化液中苯乙酮和肉桂酸的含量 被引量:11

摘要 采用等吸收点双波长紫外分光光度法,对以肉桂酸为底物,微生物转化合成苯乙酮转化液中的肉桂酸和苯乙酮,并进行直接定量分析。选择菌体发酵液作参比,检测波长分别为249nm和298nm。苯乙酮的检出限为0.22μg·mL-1(S/N=3),肉桂酸的检出限为0.24μg·mL-1(S/N=3)。两者测定结果的平均相对标准偏差分别为0.13%和0.24%;平均回收率为104.17%和97.92%。 A method for determination directly of acetophenone and cinnamic acid in the bioconversion broth of acetophenone from cinnamic acid by dual-wavelength UV spectrophetometry was developed.The fermentation broth was used as reference solution and the wave-lengths selected were 249nm and 298nm.The results showed that the detection limits of acetophenone and cinnamic acid were 0.22μg.mL^-1 and 0.24μg.mL^-1 ( S/N = 3).Both determination results of the relative standard deviation were 0.13% and 0.24%, and the average recoveries were 104.17 % and 97.92%, respectively.
