基于S2细胞建立钙库调控钙离子通道特异分子的筛选模型

Drosophila Schneider 2 Based Cellular Assay Model for Screening Special Molecules Against Store Operated Calcium Channels

摘要 应用FlexStation胞质溶胶钙离子检测技术,研究利用果蝇S2细胞建立钙库调控钙离子通道SOC的特异分子的筛选模型.通过三种已知钙通道拮抗剂的检测,发现培养温度为24℃,细胞密度为2×106个/mL时,利用终浓度为2μmol/L的SOC通道激动剂TG可建立稳定的细胞筛选模型. A Drosophila Schneider 2 cell based model system is tested for screening special molecules against store operated calcium channels, by using FlexStation to monitor intracellular free Ca^2+. The results of the experiments using three known calcium channel blockers to examine the model system indicated that under 24从不, no CO2 and 2× 10^6 cells/mL culture conditions, the S2 cell model is stable and convenient for high throughput drug screen after 2 μmol/L thapsigargin treatment.
