三维培养技术及其在肿瘤研究中的应用

摘要 传统的单层平面培养技术在肿瘤体外细胞模型建立、肿瘤细胞生物学行为分析中一直发挥着主导作用,但限于单层平面培养方法本身及肿瘤细胞传代次数的制约,细胞的形态、极性、基因表达乃至功能状态与体内肿瘤细胞的实际病理生理状况经常相距甚远,而基于两维培养的肿瘤细胞模型在肿瘤分子病理机制探讨中也开始受到越来越多的挑战。近些年,三维培养技术的发展及对肿瘤细胞生物学特性研究的深入,使得模拟体内组织细胞生长环境方面具有明显优势的立体培养,在肿瘤细胞生物学特性解析、药物疗效的评估中受到普遍的关注。
