阿托伐他汀对巨噬细胞ABCA1表达的影响

Effects of Atorvastatin on the Expression of ABCA1 in THP-1 Macrophage

目的观察阿托伐他汀对THP-1巨噬细胞三磷酸腺苷结合盒转运体A1(ABCA1)表达的影响。方法THP-1细胞经160 nmol/L佛波酯(PMA)孵育24 h,诱导分化成巨噬细胞,分别与不同浓度的阿托伐他汀(0、1、10、100μmol/L)作用不同时间(0 h、6 h、12 h、24 h)。提取各组细胞总RNA和蛋白质,分别采用RT-PCR和W esternB lot检测ABCA1的mRNA和蛋白的表达。结果与对照组比较,不同浓度的阿托伐他汀与巨噬细胞孵育不同时间组的细胞ABCA1 mRNA和蛋白表达无统计学差异(P>0.05)。结论阿托伐他汀对巨噬细胞ABCA1的表达无影响。

Objective To examine the effects of atorvastatin on the expression of ATP- Binding Cassette Transporter A1 （ABCA1） in THP - 1 macrophage. Methods After incubated with 160 nmol/L PMA for 24 h,THP- 1 was induced to macrophage. The macrophage was incubated with different concentration atorvastatin （0,1,10,100 μmol/L） for different time （0h,6h , 12h, 24h）, respectively. Total RNA and protein of macrophage were extracted. The levels of ABCA1 mRNA and protein were measured by reverse transcription polymerase chain reaction （RT- PCR） and Western blot. Results RT - PCR and Western Blot demonstrated that atorvastatin had no effects on the expression of ABCAI mRNA and protein in macrophage at different concentrations and time, compared with control group （P 〉0.05）. Conclusions Atorvastatin couldn＇t affect the expression of ABCA1 mRNA and protein in macrophage.
