霍乱弧菌新类型TCP氨基酸序列生物信息学分析

摘要 目的研究4种新类型霍乱弧菌(VC)毒力协同调节菌毛(TCP)氨基酸序列进行生物信息学特征。方法基因序列及氨基酸序列利用DNAStar和BLAST进行分析。结果对4种新类型tcpA推测的氨基酸序列的α螺旋、β折叠、疏水性、抗原表位等特征进行分析,并与国外发现的4种类型比较,N末端约100个氨基酸序列的抗原表位等特征高度一致,而C末端约60个氨基酸序列的抗原表位等特征差别较大。在对tcpA基因扩增及测序时,扩增片段包括tcpB基因5#端部分基因及其启动子区,序列分析表明,不同类型tcpB的启动子区差别较大,不同类型tcpB5#端部分基因存在多个位点突变,但均未导致氨基酸变异。结论4种新类型TCP和国外发现的4种类型的C末端抗原表位差别较大,保护性抗原表位主要位于C末端,提示不同类型TCP抗原交叉保护性较弱。 Objective To conduct bioinformation anlysis of amino acid sequences of 4 new types of toxin coregulated pilus (TCP)of Vibrio cholerae. Methods The gene and amino acid sequences were analyzed with DNAStar and BLAST software. Results Alpha regions,beta regions,hydrophilicity,flexibile regions,antigenic epitopes and other chracteristics of 4 new types of TCP were predicted. The 4 new types were compared with 4 types found by other authors,antigenic epitopes and other ehracteristics about 100 amino acid at N end part were similar highly,antigenic epitopes and other chracteristics about 60 amino acid at C end part were different. Promoter seuqences of different tcpB genes were different. There were some different sites at 5" part of tcpB gene,but amino acid sequences predicted were same. Conclusions Antigenic epitopes of 4 new types and 4 other types of TCP at C end part varied relatively greatly,protective antigenic epitopes were at C end part,so cross protective reactions of 8 types of TCP were weak.
