摘要
目的探讨Toll样受体2和4(Toll-like receptor,TLR2、TLR4)的表达在生殖器疱疹发病机制中的作用。方法采用流式细胞术检测32例生殖器疱疹患者及22例正常人外周血淋巴细胞、单核细胞和粒细胞表面TLR2和TLR4的表达。结果TLR2和TLR4在外周血单核细胞上的表达相对较高,而在淋巴细胞和粒细胞上的阳性率平均值均低于5%;患者和对照组相比,各细胞表面TLR2的表达水平无显著差异(P>0.05),TLR4在两组淋巴细胞上表达水平无显著差异(P>0.05),患者TLR4在单核细胞上的表达水平为(18.49%±10.01)%,显著高于对照组(8.02%±5.37)%(P<0.001);患者TLR4在粒细胞细胞上的表达水平为(1.90%±1.46)%,比对照组的(0.45%±0.25)%明显升高(P<0.001)。结论单核细胞和粒细胞表面的TLR4在对病毒识别、增强特异性的信号传导、启动抗病毒的固有免疫反应中起着重要作用。
Objective To investigate the significance of expression Toll-hke receptors in patients with genital herpes. Methods The expression of TLR2 and TLR4 on lymphocytes, monocytes and granulocytes was determined by flow eytometry in 32 patients with recurrent genital herpes and 22 normal controls. Results The expression level of TLR2 and TLR4 on monocytes was relative high, The positive rates of TLR2 and TLR4 on lymphocytes and granulocytes were a little lower (5%) in both groups. There were no significant difference in levels of TLR2 between patients and controls(P〉0.05). The Expression of TLR4 on monocytes was significantly higher in patients( 18.49%±10.01% )than that in contruls(8.02%±5.37%)(P〈0.001 ) and the positive rate of TLR4 expresssed on granulocytes also was increased in patients (1.90% ± 1 .46% ),without showing significant difference on lymphocytes between patient and controls (P〉0.05). Conclusions TLR4 on monocyte and granulocyte may play an important role in recognizing virus,enhancing specific single transduction,initiating antiviral inherent immune response in human body.
