摘要
目的观察重型再生障碍性贫血(简称SAA)患者外周血CD4+CD25+调节性T细胞转录因子FOXP3的表达水平及白细胞介素10(IL-10)的分泌水平。方法选用6例初发SAA、6例免疫治疗后造血恢复的SAA患者及8例健康对照的外周血标本。免疫磁珠分选CD4+CD25+T细胞,分别用ELISA法和RT-PCR法检测CD4+CD25+T细胞培养后IL-10分泌水平和转录因子FOXP3mRNA的表达水平。结果初发SAA组IL-10分泌水平和转录因子FOXP3mRNA的表达水平均显著低于造血恢复组和正常对照组(P<0.01),后2组间无明显差异。结论IL-10和转录因子FOXP3与SAA的发病可能有关。
Objective To investigate the expression of forkhead box protein P3 (Foxp3)and the level of interleukin-10 secreted by CD4^+CD25^+ regulatory T cell in peripheral blood of patients with severe aplastic anemia (SAA). Methods Enzyme linked immunosorbent assay(ELISA)and reverse transcription polymerase chain reaction (RT-PCR)were separately used to examine the level of IL-10 and expression of Foxp3 mRNA in peripheral blood of six newly diagnosed SAA patients, six recovered SAA patients and eight normal controls. Results The level of IL-10 and expression of Foxp3 mRNA in newly diagnosed SAA patients was significantly lower than those of the recovered SAA patients and normal controls,without significant difference between recovered SAA patients and normal controls. Conclusion The level of IL-10 and expression Foxp3mRNA might be correlated with the pathogenesis of SAA.
