摘要
目的:探讨姜黄素对大鼠HSC-T6表达PDGF-BB、PDGFRβ、ERK1的影响。方法:采用MTT法观察不同质量浓度姜黄素对HSC细胞增殖的影响,并计算其抑制率,确定姜黄素最佳等比浓度,设对照组、TGF-β1刺激组和姜黄素各质量浓度干预组。采用免疫组化、聚合酶链式反应(RT-PCR)半定量法探讨姜黄素对HSC-T6PDGF-BB、PDGFRβ、ERK1表达的影响。结果:正常培养的HSC-T6均有PDGF-BB、PDGFRβ、ERK1表达,经姜黄素不同质量浓度干预后,PDGF-BB、PDGFRβ、ERK1表达降低,且有一定的剂量依赖性。结论:姜黄素能抑制HSC-T6PDGF-BB、PDGFRβ、ERK1的表达,这可能是其抗肝纤维化的作用机理。
Objective:To observe the effects of curcumin on expression of PDGF-BB,PDGFRβ and ERK1 of rat Hepatic stellate cell(HSC-T6). Methods:The cultured HSC were divided into control group,each curcumin treating group. The influence of cureumin on HSC proliferation was detected by MTT assay. The protein levels of PDGF-BB , PDGFRβ and ERK1 was detected by immunhistochemical examination;The influence of curcumin on expression of PDGF-BB, PDGFRβ and ERK1 mRNA was detected by RT-PCR. Resuhs:Immunohistiochemical results showed that normal cultured HSC-T6 cell expressed PDGF-BB, PDGFRβ and ERK1 remarkably. After the intervention of different concentration of curcumin,the masculine degrees of PDGF-BB,PDGFRβ and ERK1 and the masculine cell population decreased obviously. The RT-PCR results indicated that the expressions of PDGF-BB, PDGFRβ and ERK1 mRNA could be obviously seen in control and TGF-β1 stimulus group. After the intervention of different concentration of curcumin, the expression of PDGF-BB,PDGFRβ and ERKt mRNA decreased in a dose-dependent manner. Conclusion:Curcumin could inhibit the expression of PDGF-BB,PDGFRβ and ERK1 which might be the mechanism of action curcumin on anti-fibrosis.
