摘要 目的:探讨黄芪注射液在生理剂量雌二醇环境下对人乳腺癌MCF-7细胞增殖与凋亡的影响。方法:实验设细胞对照组,他莫昔芬对照组和生理剂量雌二醇环境下5个不同剂量的黄芪注射液组。用MTT法检测细胞增殖,流式细胞仪分析细胞周期,PI染色检测凋亡率,DAN ladder观察凋亡条带。结果:各剂量组黄芪注射液均无促MCF-7细胞增殖效应,且随作用时间延长,均能抑制MCF-7细胞增殖(P<0.05),并呈剂量依赖性。在质量浓度为2×10-1~2×10-4g/mL时,各剂量黄芪注射液作用24 h均可使MCF-7细胞周期阻滞于G0/G1或S期(P<0.05)。高剂量(2×10-1g/mL)黄芪注射液作用72h可诱导MCF-7细胞凋亡。结论:一定剂量范围内的黄芪注射液可抑制激素受体阳性乳腺癌细胞的增殖,并诱导其凋亡,阻滞细胞的有丝分裂于G0/G1或S期。 Objective:To investigate the effect of Astragalus mongholicus injection on proliferation and apoptosis in hormone sensitive breast cancer cell lines(MCF-7) with physiological dose E2. Methods:Cell control group,TAM control group and five different dose Astragalus mongholicus injection groups with physiological dose E2 were set in the study. Proliferation of MCF-7 cells were evaluated with MTT assay, cell apoptotic rate were measured with flow cytometry, DNA ladder and cell cycle were observed. Results:In physiological dose E2 ,Astragalus mongholicus injection inhibited MCF-7 cells proliferation at all concentration groups. As time lasting,Astragalus mongholicus injection showed better inhibitory effect than TAM (P 〈 0. 05). Among 2 × 10^-1 g/mL -2 × 10^-4 g/mL concentration,Astragalus mongholicus injection significantly increased the proliferative percent of G0/G1 and S-phase cell, decreased percent of G2-M phase cell(P 〈 0. 05) at 24 hours. After cocuhured 72 hours,Astragalus mongholicus injection increased the rate of apoptosis to 16. 7% at 2 × 10^ - 1 g/mL concentration. Conclusion : In some dose limit, Astragalus mongholicus injection may inhibit proliferation, induce apoptosis and interrupt caryocinesia at G0-G1 phase or S phase in hormone sensitive ( MCF-7 ) breast cancer cell lines with physiological dose E2.
