虫草多糖含药血清对TGF-β_1诱导的肾小球系膜细胞增殖及Ⅳ型胶原表达的影响 被引量:7

摘要 目的观察虫草多糖(CP)含药血清对TGF-β1诱导的肾小球系膜细胞增殖及Ⅳ型胶原表达的影响,探讨CP治疗慢性肾衰肾小球纤维化的机制。方法结合血清药理学方法制备CP含药血清样品,用TGF-β1诱导的肾小球系膜细胞增殖,试验分空白对照组、对照组、高、中、低浓度5组,MTT法检测HMC的增殖及抑制率,ELISA法检测正常人肾小球系膜细胞株(HMC)中Ⅳ型胶原表达。结果CP含药血清均可明显抑制TGF-β1诱导的HMC增殖,含药血清可明显抑制TGF-β1诱导的HMC中Ⅳ型胶原的表达,以高、中浓度组最显著(P<0.01)。结论CP含药血清可明显抑制TGF-β1诱导的肾小球系膜细胞增殖及Ⅳ型胶原表达,这可能是CP治疗慢性肾衰的机理之一。 Purpose Research on effect of cordyceps polysaccharide serum containing on the proliferation and type Ⅳ collagen secretion in human mesangial cells(HMC) induced by TGF-β1. To explore the therapeutic effects of cordyceps polysaccharide(CP) on chronic renal failure(CRF). Methods With the method of blood serum pharmacology, HMC were induced by TGF-β1, and 5 groups were observed:blank, control serum, low, medium and high concentration serum which contained CP. MTT method was adopted to detect the proliferation and inhibition ratios of HMC. Type Ⅳ collagen production by cultured HMC was examined using an enzymelinked immunosorbent assay. Results Serum different concentrations of containing CP could inhibit the proliferation and type Ⅳ collagen production of HMC induced by TGF-β1 . The effect of high concentration serum was the most notable( P 〈 0.05). Conclusion The serum containing CP can inhibit the proliferation and the expression of type Ⅳ collagen in human mesangial cells induced by TGF-β1.
