缬沙坦对ox-LDL诱导的人ICAM-1表达的影响 被引量:1

摘要 目的:检测经氧化低密度脂蛋白(ox-LDL)干预后内皮细胞细胞间粘附分子-1(ICAM-1)mRNA的表达观察缬沙坦(Valsartan)对ICAM-1 mRNA表达的影响。方法:体外培养人脐静脉内皮细胞生长到融合状态时加入ox-LDL,对照组不加任何干预,作用24小时后,分别加入不同浓度的缬沙坦继续培养24小时,测定ICAM-1 mRNA的表达。结果:Ox-LDL可诱导内皮细胞ICAM-1 mRNA的表达,不同浓度的缬沙坦可抑制ox-LDL诱导的ICAM-1 mRNA的表达(P均<0.05),并里浓度依赖性。结论:缬沙坦抗炎及稳定动脉粥样硬化斑块的作用可能与抑制内皮细胞ICAW-1的表达有关。 Objective: The aim of this study was to investigate the effects of Valsartan treatment on Intercelluar Adhesion Molecule- 1(ICAM-1) in cultured human umbilical vein endothelial cells(HUVECs) with the ox-LDL. Methods: HUVECs were cultured in endothelial basal medium. Cells were grown to confluence and used for ox-LDL,while control group was not treated. After 24 hours, HUVECs were cocultured with the ox-LDL and Valsartan in different concentration for another 24 hours. Expression of ICAM-1 mRNA were detected. Results: Expression of ICAM-1 mRNA were detected in ox-LDL-induced HUVECs, Valsartan dose-dependently decreased the expression of ICAM-1 mRNA in ox-LDL-induced HUVECs(P 〈 0.05). Conclusions: Valsartan had the ability of anti-inflammatory and plaque stabilization which be connected with the inhibition of the expression of ICAM-1 mRNA.
