水蛭素抑制体外模拟增生性瘢痕模型的实验研究 被引量:11

摘要 目的:研究水蛭素对体外人增生性瘢痕模型的收缩能力,成纤维细胞的增殖、胶原分泌等功能的影响。方法:体外培养人增生性瘢痕成纤维细胞(HSFB),建立含增生性瘢痕成纤维细胞的胶原网架系统(FPCL),用1、10、50U/ml的水蛭素溶液干预该模型,测定凝胶块收缩指数,利用MTT染色计数活细胞,逆转录多聚酶链式反应(RT-PCR)法检测细胞Ⅰ、Ⅲ型前胶原mRNA表达。结果:浓度为10U/ml和50U/ml水蛭素溶液能够抑制HSFB的活力,使FPCL收缩指数降低,抑制FPCL中HSFBⅠ、Ⅲ型前胶原mRNA的表达(P<0.05)。结论:水蛭素溶液可抑制体外人增生性瘢痕三维模型的收缩功能和成纤维细胞增殖、分泌胶原功能。 Objective: To study the effect of Hirudin on contractibility of hypertrophic scar model, fibroblasts' proliferation and secretion in vitro. Methods: Human hypertrophic scar fibroblasts(HSFB) were cultured in vitro, and fibroblast populated collagen lattice (FPCL) was set up. The effect of Hirudin on HSFB model with exposure to 0U/ml, 1U/ml, 10U/ml or 50U/ml Hirudin for 24, 48 or 72 hours in vitro were studied. We detected the cells' vitality and proliferation by MTT method, measured the gels' contractile index and assessed the expression of Ⅰ, Ⅲ procollagen mRNA by reverse transcription PCR. Results: Hirudin could inhibit fibroblasts' proliferation and vitality, decrease the contractile index of FPCL and diminish the expression of Ⅰ , Ⅲ procollagen mRNA.Conclusions: Hirudin has a distinguished inhibitory action to proliferation, secretion and contraction of HSFB in vitro, which may be the mechanism of inhibiting hypertrophic scar.
