氟伐他汀对冠心病患者单核细胞ABCA1表达的影响

摘要 目的:以冠心病患者外周血单核细胞为研究对象,观察氟伐他汀对体外培养的冠心病患者外周血单核细胞三磷酸腺苷结合盒转运体A1(ABCA1)表达的影响。方法:于冠脉造影时无菌采集血液标本,肝素抗凝,密度梯度离心法分离患者外周血单核细胞,按作用时间与浓度分组,分别与10.0μmol/L氟伐他汀作用0h、6h、12h、24h,以及0、1.0、10、100μmol/L浓度的氟伐他汀作用24h。提取各组细胞总RNA,逆转录多聚酶链反应检测ABCA1 mRNA的表达。结果:与对照组比较,10.0μmol/L氟伐他汀作用于单核细胞6h、12h、24h以及不同浓度的氟伐他汀与单核细胞孵育24h均不能增强单核细胞ABCA1 mRNA(P>0.05)。结论:氟伐他汀不能增强冠心病患者单核细胞ABCA1的表达。 Aim The study was designed to examine the effects of fluvastatin on the expression of ABCA1 mRNA and protein in monocytes of patients with coronary heart disease in vitro. To evaluate the effect of fluvastatin therapy on antiarteriosclerotic. Methods Monocytes were isolated by density gradient method from blood of patients with coronary heart disease, then divided into different group. Different group monocytes incubated with 10. 0μmol/L flivastatin for 0h.6h, 12h.24h, or incubated with 0.1.0. 10.100μmol/L flivastatin for 24h, respectived. Total RNA of monocytes were abstracted by TRIzol reagent. The levels of ABCA1 mRNA were measured by reverse transcription polymerase chain reaction (RT-PCR). Results RTPCR demonstrated that fluvastatin couldn't increase the expression of ABCA1 mRNA in monocytes at different concentrations and times. (compared with control group:P〉0. 05). Conclusions Fluvastatin couldn't increase ABCA1 mRNA and protein expression in monocytes.
