摘要
目的探讨苍白球微量注射5-羟色胺(5-HT)对帕金森病模型大鼠旋转行为的调节作用。方法大鼠右侧内侧前脑束注射6-羟多巴胺(6-OHDA)以制备帕金森病大鼠模型,2周后大鼠损毁侧苍白球埋置套管,恢复3 d后苍白球微量注射5-HT,观察5-HT对帕金森病模型大鼠旋转行为的影响。结果苍白球微量注射5-HT(0.1 mmol/L)不能使大鼠产生明显的旋转行为及姿势改变。腹腔注射阈下剂量左旋多巴(L-DOPA,3 mg/kg),大鼠仅产生微弱的对侧自发性旋转行为,与5-HT组相比,差异无显著性(P>0.05)。而苍白球微量注射5-HT联合腹腔注射L-DOPA则可以明显增加大鼠的对侧自发性旋转圈数,与5-HT组相比,差异有显著性(u=3.00,P<0.05);与阈下剂量L-DOPA组相比,差异亦有统计学意义(u=16.00,P<0.01)。结论5-HT可能通过调节苍白球的兴奋性来增强L-DOPA的抗帕金森病效应。
Objective To investigate the effects of intra-pallidal microinjection of 5-hydroxytryptamine (5 HT) on rota- tional behavior in rats. Methods A model of Parkinson disease was constructed by injecting 6-OHDA into the right medial fore brain bundle of rats. Two weeks later, a guide cannula was implanted into the globus pallidus (GP) of the destructed side. After three day's recovery, 5 HT was microinjected into GP and the effects on rotational behavior was observed. Results Intra pallidal injection of 5-HT (0.1 mmol/L) alone did not induce any rotational behavior or postural changes. Intraperitoneal injection of a subthreshold dose of 3,4 Dihydroxy-L-phenylalanine (L-DOPA, 3 mg/kg i. p. ) could induce only a mild contralateral rotational behavior, compared with 5-HT, no significant difference was noted (P〉0.05). Combined intra-pallidal injection of 5-HT and intra peri toneal injection of L-DOPA dramatically increased the number of opposite-side spontaneous rotations (u= 3.00, P〈0.05 vs. 5 HT; u= 16.00,P〈0.01 vs. L-DOPA). Conclusion Anti-Parkinson effects of L DOPA probably through regulating the excitability of globus pallidus by 5-HT.
