盐酸氨基胍对糖基化终产物诱导的人肾小球系膜细胞调节活化蛋白表达的影响 被引量:2

Effects of Aminoguanidine on RANTES Expression Induced by Advanced Glycosylation End Products in Human Renal Mesangial Cells

摘要 目的探讨盐酸氨基胍对糖基化终产物(AGEs)促进人肾系膜细胞(HRMC)趋化因子RANTES表达和分泌的干预效应。方法常规方法制备糖基化终产物(AGEs-BSA)和盐酸氨基胍干预体外培养的HRMC,收集培养上清和细胞,ELISA法检测上清中RANTES浓度,实时定量RT-PCR检测RANTES mRNA表达水平,western blot检测RANTES蛋白表达。结果盐酸氨基胍干预组HRMC RANTES的mRNA和蛋白表达以及蛋白分泌明显低于AGE-BSA组(P<0.05);且降低水平呈浓度依赖性。结论盐酸氨基胍能抑制AGEs诱导的HRMC RANTES的表达和分泌。 Objective To investigate the effects of Aminoguanidine on the expression of chemokine RANTES induced by advanced glycosylation end products(AGEs) in the cultured human renal mesangial cells (HRMC). Methods AGE-BSA was prepared by incubation of bovine serum albumin(BSA) with glucose at 37℃ for 12 weeks. HRMC was cultured in the presence of AGE-BSA(glucose at 50 mmot L^-1) together with aminoguanidine. RANTES mRNA was analyzed by quantitative real time RT-PCR. The contents of RANTES in the cultured supernatant and cell lysates were detected by ELISA and Western blotting, respectively. Data were expressed as the mean ± standard deviation. Comparisons among groups were done by one-way ANOVA. Results Both mRNA and protein expression and secretion of RANTES in the HRMC treated with aminoguanidine were significantly lower than those of AGE-BSA group. The decrease of RANTES expression in aminoguanidine group was in dose dependent manner. Conclusion Aminoguanidine could inhibit the expression and secretion of RANTES induced by AGE-BSA.
