摘要
目的建立一种适合膜片钳研究的成年大鼠背根神经节(Dorsal root ganglion,DRG)细胞培养方法。方法分离并培养成年大鼠DRG神经元,显微镜下观察培养神经元的形态,膜片钳技术记录培养神经元的基本膜电生理特性及ATP激活电流。结果培养的DRG神经元形态结构完整;其静息膜电位在正常范围内,可记录到诱发动作电位及钠电流;在膜片钳全细胞记录模式下,能稳定地记录到ATP诱发电流。结论培养的成年大鼠DRG神经元适合膜片钳试验。
Objective To establish a kind of cultural method of dorsal root ganglion (DRG) neurons for patch clamp study. Methods DRG neurons of adult rat were isolated and cultured. The electrophysiological properties and ATP-induced currents of cultured DRG neurons were investigated by using patch-clamp techniques. Results The cultured cells were morphologically intact, from which evoked action potential and Na^+ channel currents, and ATP-induced currents could be detected. Conclusions Cultured DRG neurons of adult rat are ideal for patch-clamp studies.
