补肾益骨方对去卵巢大鼠骨质疏松骨密度及IL-6,TNF-α的影响

Effects of bushen yigu recipe on bone mineral density and blood level of interleukin-6,tumor necrosis factor-α in ovariectomized rats

目的探讨中药补肾益骨方对去卵巢大鼠骨质疏松骨密度及IL-6、TNF-α的影响。方法运用切除大鼠卵巢方法建立去卵巢骨质疏松症模型,随机分为对照组、模型组、倍美力组、补肾中药组。采用双能X线骨密度测定法检测各组实验鼠骨密度(BMD);制备各组大鼠腰椎石蜡切片,观察各组大鼠腰椎骨组织形态学变化;采用ELISA法及放射免疫分析法检测各组实验鼠血清抗酒石酸酸性磷酸酶5b(TRACP5b)、白细胞介素-6(IL-6)、肿瘤坏死因子(TNF-α)。结果补肾益骨方可明显升高BMD,降低TRACP5b、IL-6和TNF-α含量,明显改善大鼠腰椎骨组织形态学。结论去势骨质疏松模型大鼠血清TRACP5b、IL-6和TNF-α明显升高,补肾益骨方可以明显降低上述血清学指标,同时明显改善大鼠腰椎骨密度及骨组织形态,进一步从分子细胞水平证实该方对绝经后骨质疏松症有很好的防治作用。

Objective To explore the effect of Bushen Yigu recipe on bone mineral density and blood level of interleukin-6,tumor necrosis factor-or in ovariectomized rats. Method The model of osteoporosis was established by ovariectomy in female rats,and then they were divided into four groups： the test group of Bushen Yigu Decoction,the conjugated estrogens group,the model control group and the blank control group. Bone mineral density （BMD） was measured by dual energy X-ray absorptiometry. And we examine the change of bone histomorphology by bone biopsy and detect the serum level of tartrate acid phosphatase 5b （TRACP5b） ,interleukin-6 （IL-6） and tumor necrosis factor-α （TNF-α） by ELISA and Radioimmunoassay. Result the BMD was higher in the Bushen Yigu Decoction group. And the Bushen Yigu Decoction can lower the level of TRACP5b,IL-6 and TNF-α. The histomorphology shows better in the lumbar vertebrae. Conclusion From the molecular and cell level,the study discovers the mechanism. The Bushen Yigu Decoction is effec- tive on decreasing the level of TRACPSb,IL-6 and TNF-α in ovariectomized rats. It can also improve the BMD and ameliorate the hisomorphology.
