摘要
目的探讨聚羟基丁酸-羟基戊酸共聚酯(PHBV)膜的生物相容性及其成骨能力。方法采用骨髓基质细胞体外培养技术,通过MTT法检测细胞相对增殖度,对PHBV的细胞毒性进行分级评价;并制备犬胫骨骨缺损模型,观察PHBV膜引导骨再生的效果。结果PHBV膜的细胞毒性分级为0~1级;PHBV膜覆盖组术后第2周即可见新骨形成,12周时骨缺损区已完全为新骨充填,骨修复质量明显优于对照组。结论PHBV膜对骨髓基质细胞无明显毒性,能引导骨组织再生,有望成为一种较理想的引导骨组织再生的天然膜材料。
Objective To study the biocompatibility of poly (hydroxybutyrate-hydroxyvalerate) (PHBV), a novel biopolymer membrane, and evaluate its efficiency of promoting bone regeneration in vivo. Methods Cell relative growth rate (RGR) was measured by MTT method to quantitatively evaluate the cytotoxieity of the biomaterials. Eight healthy dogs were used as experimental animals. Four bone defects were made on bilateral tibias in each dog. The PHBV-membrane-guided bone generation effect was assessed by histological observation. Results The toxicity gradation of PHBV membrane ranked from grade 0 to grade 1; new bone regeneration was observed in the defects covered with PHBV membranes at 2^nd week post-surgery. The defects covered with PHBV membranes were filled with mature bone at 12^th week post-surgery. The bone rehabilitation in experimental groups was earlier and better than that in control group. Conclusion PHBV membrane, which has no cytotoxicity to MSCs in a wide range of extract concentration, may be applied as a promising biopolymer membrane for guided bone regeneration.
