摘要
目的探讨姜黄素对体外培养人舌鳞癌Tca8113细胞增殖和凋亡作用。方法不同浓度的姜黄素处理体外培养的人舌鳞癌Tca8113细胞,MTT法检测细胞增殖,流式细胞仪检测细胞凋亡,并行凋亡相关蛋白Fas、P53、Bcl-2的免疫组化测定。结果在10、30、50、70μmol/L浓度范围内,姜黄素可剂量依赖性抑制人舌鳞癌Tca8113细胞增殖的作用。30、40、50μmol/L三个不同浓度姜黄素处理组的凋亡指数与对照组比较差异有统计学意义(P<0.05),细胞凋亡率与药物浓度正相关。30、40、50μmol/L不同浓度姜黄素处理组Fas、P53、Bcl-2凋亡细胞阳性率与对照组比较差异具有统计学意义(P<0.05)。结论姜黄素可显著抑制人舌鳞癌Tca8113细胞增殖,并诱导其凋亡,其作用呈剂量依赖关系。
Objective To inverstigate the effect of curcumin on proliferation and apoptosis of human tongue carcinoma cell line Tca8113. Methods Human tongue carcinoma cell line Tca8113 was incubated with different concentrations of curcumin, cell proliferation was examined by MTT colormetric assay. Apoptosis was analyzed by flow cytometry (FCM), and apoptosis related protein Fas, p53, bcl-2 were examined by immuno-histochemical staining. Results Administration of 10, 30, 50, 70 μmol/L curcumin could significantly inhibit human tongue carcinoma cell line Tca8113 in a dose-dependent manner. After the cells being treated with curcumin at different concentration (30, 40, 50 μmol/L) for 24 hours, the apoptosis indexs (%) were (55.25 ± 4.78), (66.34 ±5.43) and (84.36 ±6.56) respectively, which were significantly higher than that of control group (6.57 ±0.74, P 〈 0.05) and in dose-depended manner.In the immunohistochemical examination, cells were treated with curcumin of different concentrations (30, 40, 50 μmol/L for) for 24 hours, the positive apoptosis cells rates of Fas, p53, bcl-2(%) were (56.25 ± 3.75, 64.72 ± 5.86, 78.64 ±8.47)%, (51.78 ± 4.58, 65.15 ± 6.02, 84.32 ± 7.65)%, (21.36 ± 3.24, 18.92 ± 2.65, 12.75 ± 2.54)% respectively, significantly higher than that of control group (3.26 ±0.32)%, (2.18 ±0.24)%, (35.54 ± 5.36)%, (P 〈 0.05). Conclusion Curcumin can significantly inhibit human tongue carcinoma cell line Tca8113's proliferation and can induce its apoptosis in dose-depended manner.
