摘要
目的研究肝窦内细胞(HSEC)与凋亡细胞互相作用后促炎症因子分泌水平的变化。方法紫外线诱导人淋巴细胞凋亡;对照组:体外人HSEC单独培养。实验组:人HSEC与同种异体凋亡淋巴细胞细胞直接共培养。两组均培养16h后,收集上清液,利用Luminex技术检测上清液促炎症性的细胞/化学因子水平,包括IL-2、ⅡNF-γ、TNF-α。结果实验组IL-2、ⅡNF-γ、TNF-α分泌水平比对照组下调约50%,两组数值的差异有显著性(P<0.05)。结论HSEC与凋亡细胞作用后能使促炎症因子下调,调控局部免疫微环境,使免疫反应偏向耐受。
Objective To investigate the effects of apoptotic lymphocytes on the secretion of cytokines by hepatic sinusoidal endothelial cells (HSEC). Methods Human HSEC cells were co-cultured for 16 h with allogenetic apoptotic lymphocytes induced by UVB irradiation. The supematants were collected and the levels of interleukin-2, interferon-γ and tumor necrosis factor-α were detected by Luminex technique. Results All the cytokines were down-regulated by about 50% in HSECs after co-culture with the apoptotic lymphocytes as compared with those in the control group (P〈0.05). Conclusion Co-culture with apoptotic lymphocytes can down-regulate the secretion of pro-inflammatory cytokines in HSECs, which may contribute to tolerogenic microenvironment in the liver.
