摘要
目的:建立同种同基因和异基因大鼠子宫原位移植模型,评价移植子宫的生殖功能。方法:同基因组以雌性Wistar大鼠分别作为供体和受体,行子宫原位移植术。异基因组以雌性SD大鼠作供体,雌性Wistar大鼠作受体,行子宫原位移植术,受体大鼠术后第1天起使用环孢素,每天每公斤体重肌肉注射10mg。对照组雌性Wistar大鼠10只,切除左侧宫角。术后3组大鼠分别与同周龄雄性Wistar大鼠合笼12周,自然交配,观察受孕及分娩情况。子代大鼠自出生起每周称量体重至第8周,绘生长曲线,并各自分别交配。结果:10例同基因大鼠,10例异基因大鼠分别行子宫原位移植手术,10例对照手术。受体大鼠存活率同基因组80%(8/10),异基因组90%(9/10),对照组大鼠存活率90%(9/10)。同基因组8例存活的大鼠中4例受孕,受孕率50%,顺产分娩子代大鼠15只;异基因组9例存活大鼠中2例受孕,受孕率22.2%,但2例均胎死宫内;对照组9例存活大鼠中6例受孕,受孕率66.7%,顺产分娩子代大鼠19只。同基因组和对照组分娩的子代大鼠外观均无畸形,两组子代生长曲线基本相似,并有正常生殖功能。结论:同基因大鼠子宫原位移植后移植子宫可恢复正常生殖功能;异基因大鼠子宫原位移植后应用环孢素可使移植子宫长期存活,但生殖功能可能受影响。
Objective:To establish the orthotopic uterine transplantation model in isogenic rats and allogeneic rats and assess the fertility of transplanted uterus. Methods: Twenty female Wistar rats were used as donor and recipient, and orthotopic uterine transplantation was conducted in the isogenic group. Ten female SD rats were used as donor and ten female Wistar rats were used as recipient, and orthotopic uterine transplantation was conducted in the allogeneic group. Cyelosporine was muscle injected from the first day after surgery in the receptor rats. The 10 female Wistar rats were resected the left uterine horn in the control group. After oprate was 80% (8/10) in the isogenic group,90% (9/10) in the allogeneic group and 90% (9/10) in the control group respectively. Out of the 8 recipient rats in the isogenic group, 4 pregnancies were achieved and 15 baby rats were delivered spontaneously. The fertility rate was 50%. Out of the 9 recipient rats in the allogenic group ,2 pregnancies were achieved but the baby rats were all dead within the womb. The fertility rate was 22.2%. In the control group, 6 pregnancies were achieved and 19 baby rats were delivered spontaneously. The fertility rate was 66.7%. All baby rats in the isogenic group and control group were no teratism of external appearance. The growth trajectories of the progeny from two groups were coincidence on the whole,and all progenies had normal fertility. Conclusion: Orthotopic transplanted uteri in the isogenic rat can restore the normal fertility. Cyclosporine can make allogeneic transplanted uteri long-term survival, but the fertility of the transplanted uteri may be affected.
