摘要 HIV-Tat蛋白转导域具有穿膜活性,能够将与之共价连接的多肽、蛋白、核酸等生物大分子快速而高效地转导入细胞内部,且对细胞无毒副作用,在药物转运及疾病治疗等领域有着广阔的应用前景。本文对HIV-Tat蛋白的结构、穿膜机制及转导域功能的应用作一综述。 HIV-Tat protein transduction domain (PTD) has the ability to cross the plasma membrane of mammalian cells. The biological macromolecules linked covalently with HIV-Tat PTD, such as polypeptides, proteins and DNAs, may be transducted into cells rapidly and effectively, without toxicity or side effect. HIV-Tat PTD is of a broad prospect in delivery of drugs and treatment of human diseases. The structure, transmembrane mechanism and application of Tat protein are reviewed in this paper.
