乳源免疫调节肽对人卵巢癌SKOV3细胞生长及凋亡的影响 被引量:5

摘要 目的:探讨乳源免疫调节肽对人卵巢浆液性乳头状囊腺癌SKOV3细胞生长和凋亡的影响。方法:不同浓度乳源免疫调节肽对体外培养的SKOV3细胞作用24、48、72及96 h后,应用MTT法检测细胞的生长抑制情况,采用HE染色、透射电子显微镜观察及FCM分析细胞周期及细胞凋亡情况。结果:乳源免疫调节肽对SKOV3细胞有增殖抑制作用,与对照组相比差异有统计学意义(P<0.05),且增殖抑制作用具有时间和浓度依赖性。HE染色和电子显微镜下均观察到用药后出现凋亡细胞的形态学特征。FCM检测发现,乳源免疫调节肽作用后细胞的凋亡率与对照组相比,差异有统计学意义(P<0.05),其中3×10-4g/L PGPIPN(Pro-Gly-Pro-Zle-Pro-Asn)组的细胞凋亡率最高,达29.4%。结论:乳源免疫调节肽可以抑制体外培养的人卵巢浆液性乳头状囊腺癌SKOV3细胞的增殖,并可诱导其凋亡。 Objective :To investigate the effects of immunommc, dulating peptide frmn β-casein (PGPIPN) on proliferation and apoptosis of ovarian papillary serous cystadenocarcinoma SKOV3 cells. Methods : SKOV3 cells were incubated with different concentrations of PGPIPN for 24, 48, 72, and 96 h. Inhibitory effect of PGPIPN on SKOV3 proliferation was tested by MTT assay. Apoptotic cells and apoptotic bodies of SKOV3 cells induced with PGPIPN were observed by hematoxylin-eosin (HE) staining and electron microscopy (EM). Cell apoptotic percentage and cell cycle distribution were measured by flow cytometric assay (FCM). IResults:PGPIPN inhibited the proliferation of SKOV3 ceils in vitro compared with control group (P 〈 0.05 ). The inhibitory effects were in a time- and concentration-dependent manners. The characteristic morphological changes of apoptosis were observed by HE staining and EM in SKOV3 cells after PGPIPN treatment. FCM analysis found that the apoptotic rate was significantly different between PGPIPN-treated and control groups. The difference was significant (P 〈 0.05). The apoptitic rate reached the maximum (29.4%) in PGPIPN 3 ×10 ^-4 g/ L treated group. Conclusion: PGPIPN inhibited proliferation and induced apoptosis of ovarian papillary serous cystadenocarcinoma SKOV3 cells in vitro.
