大豆甙元对去卵巢大鼠雌二醇、维生素D和骨矿沉积的影响 被引量:3

Effects of daidzein on the expressions of estradiol and 25-OH-D and bone minerals deposition in ovariectomized rats

摘要 目的研究不同剂量大豆甙元对去卵巢大鼠雌二醇(E2)激素水平和骨矿沉积的影响。方法60只3月龄雌性大鼠分为6组:对照组、模型组、4个去卵巢+大豆甙元组,每日分别灌服10 mg/kg蒸馏水和25、50、75、100 mg/kg大豆甙元;9月龄时处死,分别测定骨灰分、Ca、P、Mg、Zn和血清中E225羟-基维生素D(25-OH-D)及骨钙素(BGP)。结果服用大豆甙元后,去卵巢大鼠血清E2水平未升高,但25-OH-D和BGP水平恢复正常,从而增加了骨Mg和Zn沉积。结论大豆甙元通过调高(25-OH-D)及BGP水平,帮助骨矿沉积,可获得与雌激素替代疗法(ERT)近似的临床效果,但不诱导雌激素高表达,使用安全性高。 Objective To evaluate the daidzein on the expressions of estradiol ( E2 ) and 25-OH-D and bone minerals deposition in ovarieetomized (OVX) rats. Methods 60 3-month-old female rats were divided into normal control, OVX and four different doses of daidzein (25, 50, 75, 100 mg/kg BW) treatment groups. At the end of trial (age of 9-month), all the rats were killed to collect their leg bone and blood as samples to test bone mineral, Ca, P, Mg, Zn in bone by atomic absorption method or Ca, P, E, 2,5-OH-D and BGP in serum by ELISA. Results The serum E2 level was not changed in OVX rats with or without daidzein. The Mg, Zn deposition in bone were in- creased because the serum levels of 25-OH-D and BGP were recovered in daidzein treatment groups. Conclusions Daidzein could improve the bone minerals deposition through up-regulating the levels of 25-OH-D and BGP, which is similar to ERT but more safety because the level of E is not increased.
