金属硫蛋白3对快速老化痴呆模型小鼠海马CA1区神经元细胞凋亡的影响 被引量:1

摘要 目的研究金属硫蛋白3(MT3)对快速老化痴呆模型小鼠(SAMP8)海马CA1区神经元细胞凋亡的影响及其机制。方法将7月龄SAMP8小鼠随机分成6组,分为痴呆组、3种浓度的MT3组、MT1组、空白对照组。快速老化小鼠对照(SAMR1)小鼠为正常对照组。连续腹腔注射28 d后灌注取脑,进行海马CA1区原位末端标记(Tunel)试剂盒和免疫组化(SP)试剂盒染色。结果痴呆组海马CA区神经元细胞凋亡指数明显高于正常对照组。高浓度MT3组凋亡指数最低,Bcl-2蛋白表达增强,Bax蛋白表达最低(P<0.01,P<0.05)。MT3组对Bax蛋白表达的抑制作用强于MT1对照组,而二组对Bcl-2蛋白表达的差异不显著。结论SAMP8小鼠海马结构存在大量神经元细胞凋亡,MT3通过减少Bax表达和增加Bcl-2表达,减少细胞凋亡,并有浓度依赖关系;且作用强于MT1。
