RNA介导ABCE1基因沉默对肺癌细胞95-D增殖、凋亡的影响

摘要 目的研究构建的ABCE1基因SiRNA表达质粒基因沉默效果,及其对95-D肺癌细胞株增殖、凋亡能力的影响。方法运用转染试剂FuGENE 6对3种不同的ABCE1基因SiRNA表达质粒在95-D肺癌细胞中进行转染,并用G418筛选,荧光显微镜观察3种质粒的转染效果,收集转染细胞,计数转染细胞总数、运用MTT法检测细胞活力,用ELISA法检测细胞凋亡。结果①转染的细胞表达红色荧光蛋白,荧光显微镜结果表明,重组质粒转染效率达42.7%。②实验组的细胞增殖低于对照组、而细胞凋亡高于对照组(P<0.05)。结论ABCE1特异性siRNA可明显抑制ABCE1基因转录和表达,并能有效诱导95-D细胞的凋亡,为下一步在体内利用Si-1重组质粒沉寂ABCE1基因,促肿瘤细胞的凋亡提供实验基础。
