摘要
[目的]考察全自动定氮仪测定小麦籽粒蛋白质的可行性。[方法]以皖麦44和烟麦19为供试品种,应用KJELTEC2300型全自动凯氏定氮仪测定小麦籽粒蛋白质。研究该测定方法的精确度和准确性,并与经典凯氏定氮法的测定结果进行比较。[结果]皖麦44全麦粉总氮量的添加回收率在99.38%~100.35%;皖麦44和烟农19的蛋白质含量10次平行测定结果的变异系数分别为O.60和0.84,符合国标方法允许误差的要求;经显著性测验表明,全自动定氮仪与经典的凯氏定氮法对皖麦44和烟农19全麦粉的蛋白质测定结果无显著性差异。[结论]用KJELTEC2300型全自动定氮仪测定小麦籽粒蛋白质的结果与经典凯氏定氮法的吻合,不仅具有很好的精确度和准确度,还具有操作简便和快速省力的特点,可广泛用于小麦籽粒的蛋白质含量测定。
[ Objective] The study aimed to investigate the feasibility of determining protein in wheat grains by fully automatic azotometer. [ Method ] With Wanmai 44 and Yannong 19 as the tested varieties, their grain protein was determined by KJELTEC 2300 Fully Automatic Azotometer to study the precision and accuracy of this determining method and its determined result was compared with that by the classical Kjeldahl method. [ Result] The total N amount in wholemeal of Wanmai 44 had the fortified recovery rate of 99.38% -100. 35%. The variation coefficient of 10 parallel determinations on protein contents in wholemeal of Wanmai 44 and Yannong 19 were 0.60 and 0. 84 resp. , which accorded the error request for national standard that could be allowed. The significant test showed that there was no significant difference of protein content either in Wanmai 44 or Yannong 19 between the full automatic azotometer method and the classical Kjeldahl method. [ Conclusion ] KJELTE C2300 Fully Automatic Azotometer in determination on the protein in wheat grains inosculated with that by the classical Kjeldahl method, and it not only had very good precision and accuracy for determination, but also had characters of simple operation, rapid and laboursaving, which could be widely applied in determining protein in wheat grains.
